Computational Identification of Phosphorylation Sites around Nuclear Localization Signal Sequence Reveals New Insight into Genes Associated With Human Diseases

چکیده
Alterations in protein Subcellular localization often contribute to the development of human diseases. Post-transcriptional modifications, such as phosphorylation on the Nuclear Localization Signal (NLS), may change the protein’s localization. However, little is known about the frequency and local effects of phosphorylation near NLS sites. In this study, a computer program was developed to search various databases in order to find proteins with NLS Phosphorylations, and any diseases that are associated with those genes. 308 NLS sequences were found in the NLSdb database, resulting in the identification of 133,448 NLS-containing proteins in the Uniprot database. We cross-referenced these proteins with phosphorylation data available from the PhosphoSitePlus database and found that about 21% of these NLS-containing proteins have evidence of phosphorylation sites. After plugging this into the gene disease association database, 138 of disease-associated genes (1% of NLS-containing proteins)were identified to have phosphorylation sites on their NLS sequences. Further evaluation of the NLS phosphorylation status of these genes in clinical samples may lead to development of new biomarkers for human diseases, and shed new light into the pathogenesis of these gene-associated diseases. *Corresponding author: Dr. Jianjun Hu, Department of Computer Science and Engineering, University of South Carolina, Columbia, SC, 29208, USA; Tel: +1(803)7777304; Fax:+1(803)7773747; Email: [email protected] Received Date: November 11, 2016 Accepted Date: December 02, 2016 Published Date: December 17, 2016

Cloning and molecular characterization of TaERF6, a gene encoding a bread wheat ethylene response factor
Ethylene response factor proteins are important for regulating gene expression under different stresses. Different isoforms for ERF have previously isolated from bread wheat (Triticum aestivum L.) and related genera and called from TaERF1 to TaERF5. We isolated, cloned and molecular characterized a novel one based on TdERF1, an isoform in durum wheat (Tri...

Today, antimicrobial peptides are known as a new generation of antibiotics for treatment of microbial diseases in human and animals and protecting plants against different pathogens. Heveins are a group of antimicrobial peptides which are considered as one of the most important groups of antimicrobial peptides due to the very high diversity and expression in different plant organs as well as th...

The nuclear presence of the expanded disease proteins is of critical importance for the pathogeneses of polyglutamine diseases. Here we show that protein casein kinase 2 (CK2)-dependent phosphorylation controls the nuclear localization, aggregation and stability of ataxin-3 (ATXN3), the disease protein in spinocerebellar ataxia type 3 (SCA3). Serine 340 and 352 within the third ubiquitin-intera...

Tyrosine phosphorylation is a dynamic reversible post-translational modification that regulates many aspects of cell biology. To understand how this modification controls biological function, it is necessary to not only identify the specific sites of phosphorylation, but also to quantify how phosphorylation levels on these sites may be altered under specific physiological conditions. Due to its...
